A Water?Soluble Peptoid Chelator that Can Remove Cu <sup>2+</sup> from Amyloid?? Peptides and Stop the Formation of Reactive Oxygen Species Associated with Alzheimer's Disease

Cu bound to amyloid-? (A?) peptides can act as a catalyst for the formation of reactive oxygen species (ROS), leading neuropathologic degradation associated with Alzheimer's disease (AD). An excellent therapeutic approach is use chelator that selectively remove from Cu–A?. This should compete Zn2+ ions (Zn) are present in synaptic cleft while forming nontoxic complex. Herein we describe P3, water-soluble peptidomimetic removes Cu2+ Cu–A? presence Zn and prevent ROS even reductive environment. We demonstrate, based on extensive spectroscopic analysis, although P3 extracts Cu,Zn–A? faster than it Cu, formed complexes kinetic products further dissociate, CuP3 an exclusive stable thermodynamic product. Our unique findings, combined bioavailability peptoids, make drug candidate context AD.
